The purpose of the current survey was evaluation of the anti-influenzal effect of the “ORVI-BIOFON” apparatus during the experimental influenza infection in non-pedigree white mice.

METHODS

For attainment of the inquiry purpose two experiment series have been staged. During the first series of the experiments the anti-influenzal effect of the “ORVI-BIOFON” apparatus was evaluated during inquiry into the lethality among white mice infected with the grippe strains virulent for these animals. In the other series of the experiments evaluation of the exposure effect of the said apparatus was evaluated through inquiry into reproduction of the grippe virus in the lungs of the mice. The experiments were carried out in non-pedigree white mice weighing 12 – 16g. The animals received standard ration and were kept in identical conditions. The experimental and the check animals were kept in different rooms. For the experimental purpose the mice were intranasally infected under light ether narcosis by viral allantois liquid and its 10-multiples dilutions. To simulate an influenza infection, grippe virus type A/Puerto Rico/8/34 (H1N1) virulent for mice was used. The experimental animals were exposed to “ORVI-BIOFON” emission immediately after infection and then every 3-4 hours day and night for 10 days. The check animals received no exposure treatment with the “ORVI-BIOFON” apparatus. The animals stayed under observation for 14 days after being infected.

Reproduction of the grippe virus in the lungs of the mice was evaluated by viral concentrations in the lungs on the 3d and the 7th day after infecting the animals by 103-105 50/0.2 ml doses. The virus quantity was determined through the mice lung suspension titers by the Reed L., Mucnch H method.(/ Amer. J. Hyg. - 1938. - Vol.27.-P.493-497 )

The obtained results

Anti-influenzal action of the “ORVI-BIOFON” apparatus on the lethality among animals during the experimental grippe infection was studied. Two groups of mice were infected with the grippe virus doses ranging from 1 to 109 (50/0.2 ml.).

I group – 30 mice (experimental group)

II group – 30 mice (check group)

Immediately after infecting and then every 3-4 hours day and night the experimental group mice were exposed to the “ORVI-BIOFON” apparatus emission under standard conditions. Each exposure session lasted 22 seconds. Exposure and observation continued for 10 days.

It became apparent that only 9 mice infected with minimal doses of total 30 in the check group stayed alive by the 10th day. At the same time only two mice died (6.7%) in the experimental group receiving exposure treatment. The survival rate thus amounted to 94,3%.

Thus it was demonstrated that that anti-influenzal effect of the “ORVI-BIOFON” apparatus prevented development of the influenzal pneumonia and reduced the grippe mortality 10 times.

A comparative appraisal of the grippe virus reproductive ability in the lungs of white mice under normal conditions and during methodical exposure to the “ORVI-BIOFON” apparatus was conducted. The viral lung concentrations in white mice were determined on the 3d and the 7th day after infecting. As it became evident with the “ORVI-BIOFON” exposure treatment administered positive reduction of the viral quantities in he lungs was registered both on the 3d and the 7th days in comparison with the check cases. With low quantity initial infection the drop in the grippe virus concentration in the lungs of the exposure mice in comparison to the check also proved effective reducing the quantities of the reproducing virus by 0,5-0,8 lg 50/0.2ml (3-6 fold reduction) by the 3d day and by 0,3-0,7 lg 50/0.2ml ( 2-5-fold reduction) by the 7th day.

Thus regular exposure of the infected mice during the simulated grippe infection to the “ORVI-BIOFON” apparatus emission permitted to achieve a pronounced reduction the quantity of the reproduction virus in the lungs in comparison to the check group mice. With initial low quantity infection which is typical for natural grippe infection use of the “ORVI-BIOFON” exposure reduces the virus reproduction concentrations 30-fold, which may be an important factor for the organism in struggle with the infection.

The obtained results served as the basis for the following conclusion about the anti-influenzal effect of the “ORVI-BIOFON” apparatus during the simulated grippe infection in the white mice:

Use of the apparatus permits to reduce the white mice mortality at least 10-fold.

Use of the apparatus reduces significantly in comparison to the check the viral concentrations in the lungs of white mice thus decreasing viral reproduction and preventing possible complications and lethal outcome.

Employment of the “ORVI-BIOFON” apparatus is most advisable with low-quantity initial infection in animals and, obviously, at early stages of natural influenzal infection, when high levels drop in the viral reproductive activity in the lungs of the infected animals is apparent.
